If you have a single polygon shapefile and you wish to assign loads based on area, you could consider using the Load Estimation by Land Use method in LoadBuilder. This will distribute loads to manholes based on the desired flow per area along with the area of the manhole's associated service area layer. See this recent, similar discussion.
