Is it possible to define a line weight by colour? I think this can be done in AutoCAD but I have no idea how to set this up in Microstation.

Hi, you can use pen tables or the plotter driver configuration files for associating a plotting width to each of MicroStation's colour. You can refer to the section "Working with Complete Designs \Print Resymbolization and Pen Tables" of MicroStation's help to get started. HTH
There is also a neat other way, use the csv remapping feature to export a V8i dgn to another v8i dgn it creates a spreadsheet where you can import all the settings of the current file say colours and set the export to set weights against the colours ...
I have used this feature for levels and fonts before it has the options for colours and weights ( turn on optional columns) it should work with colours to weights too.

pentables only work on prints, csv remapping also works on graphic elements and prints...both methods are valid but not everyone is aware of csv remapping a DGN file in and a DGN file out ... its often thought only for format conversion ie DWG to DGN ...
Also giving a client DGN files as well as pdfs or paper... I believe its far better to use WYSIWG unlike our older ACAD users some who still print by colour both for weight and linetypes and need to provide pentables just for those odd files.....seems silly when it comes to mstn....
even though acad has had wysiwyg and weights available for several years.... ( 2007 I think)
