I am looking to inquire what the difference between the "RC-Design" type load combination and the "Design" type load combination is when designing reinforced concrete beams and columns. I have run both types and cannot figure out the difference between the two types of combinations. I could not find the difference in the manual either. Both types of combinations allow the Concrete Module.

"RC-Design" combinations are used only for the design of concrete beams, columns, walls, and foundations. If the model contained members with other any other material, the "RC-Design" combinations would not be used to design those members.
"Design" combinations are used for the design of all members regardless of material type.
If your model only contained concrete members, then you would only need to define "RC-Design" or "Design" combinations but not both.
